Not me, but take a look at this:

That's a pretty awesome mission, I wonder why he disabled ratings and comments on that vid, cos I wanted to give it a like! (as I'm sure others would have).

Most points I've got in a single mission was my 3rd and final career mission which brought back 8646.3 points.

Something above 3000 for the usual multi-landing Minmus spiel (Some data was transmitted, pretty sure I had close to zero science when the mission started):

It's sort of boring. Only interesting bit was when I undocked the lander, noticed I had forgotten to refuel, looked at the DV left, thought "screw it, that's enough"... it wasn't really. Made it back to orbit fine, but the entire rendezvous had to be done on RCS.

Some missions will bring in boatloads of science. Jool system exploration is one good example.

Rover on Mun or Minmus is another, this can be expanded if you return to mothership in orbit to refuel and land on other biomes.

Downside is that many of the huge missions is that it might often be more effective to do multiple small missions, return the science and build a more capable ship the next time.

I'm conducting a Grand Tour at the moment, it uses the maxed out tech tree, and several mods, AND is done in 0.22, AND it uses planet factory, but with just visiting Kerbin, Mun, Minmus, Eve, and Gilly, the old science overview mod estimates about 22000 science over the 300 experiments already taken on the ship and it's five landers.
