Do you need signal when you have crew?

With a pilot you have full control.

Without a pilot but with a Kerbal of another kind and a probe core, you will have all pilot abilities as if the probe was a Kerbal pilot, minus the ability to make and delete maneuver nodes.

If you only have a probe core, you will only be able to toggle SAS, aim at whatever the probe core can aim at (prograde, radial, those buttons by the navball), and go full or no throttle.

This is assuming default normal settings.

Just wanted to add that this is actually documented in the KSPedia, e.g. on page 137 on the pdf version here.(*)

You can also see which kind of control you have by looking at the icons in the upper left corner of the flight screen (which are explained on page 136 of the KSPedia document).

(*) As of 7. June 2020. IIRC SQUAD does update the document when they update the KSPedia.
