Let's say I check goo and it tells me keep = 40, transmit = 20. I transmit and get 20. When (if) I land on Kerbin and recover ship do I get the other 20?

No. When you transmit, it eats the canister.

The Goo can (And materials bay) become inoperable after transmitting (in .23) However, if you have a Mobile Lab you can process the data to get more Transmission value, and clean to goo for reuse, also, your Kerbal can EVA to the Goo Can and take the data and put it in his pod so that you only have to bring the pod back.

Hey Larry. The only way to get 100% return on goo or material experiments is to "take data" from the container or canister, store the data in a command pod and then return to Kerbin and recover the pod.

This is true for all experiments. If you transmit the data you no longer have that data to return. However, only the Goo Can and Material Bay are inoperable after transmission. Also there is a 40% cap on the amount of science you can gain by transmission (Unless the transmission value is greater than that, in which case you get the amount from one transmission, but no more) It is best to return them if you can.

Some experiments, like crew reports or the magnetometer and Radio Plasma Wave Sensor experiments from the DMagic science pack, will give you full value for transmission. But that's the exception, rather than the rule.

Also, the Kerbal Dust Experiment (KDEX) will give you 90% after a run through the lab, so it's almost worth transmitting.
