Is there a way to change some difficulty settings after a game has been started?

For instance if I want to add some difficulty later in my career and want to add the G -effects options later on?

Do you know if "Crew Respawn Timer" is real life hours, or in-game hours?

I want to say real life.  I think the scale for in-game time would probably be measured in days.
