2 things immediately come to mind.

  1. You have somehow altered the mapping of the keys that control throttle.  Unlikely, but it wouldn't hurt to look to make sure you have Z/X for full up/total kill, and then Left Shift/Left CTRL for smaller up/down increments.
  2. You are flying a probe, or a command module without a pilot, that does not have communication back to KSP or to a probe/satellite/command module that has CommNet connectivity.  I believe "Require Connection For Control" and "CommNet" are enabled by default in the settings.  This is the more likely scenario here, but you can always go into the settings and turn both of these OFF (deselect them) and you should get connectivity back and be able to fly.

If neither of those is the issue, then I'll echo what Vanamonde said above and ask for pictures of the craft.

Here's some tips.

1. Check your keybinds. You may have switched the throttle keybinds around. For reference the throttle keybinds are usually like this.

Z/X: Full and empty throttle.

LShft/LCtrl: Small throttle adjustments.

2. Check your connection to Kerbin. If Commnet and Require Connection for Control is enabled, you need antennas to go to even the Mun.

3. Check your fuel flow. If you have something blocking the fuel tanks from getting fuel to your engine, that's a big problem.

4. Verify Integrity of Game Files. If you got KSP from steam, go to the properties tab and select "Verify Integrity". This will identify issues and recompile the game's files.

5. Check if any mods are interfering. Some mods don't work well with others.

If all else fails, Re-install the game.
