At that point the craft was descending, so it was likely moving down through very hot air. Could easily be just something like grease or glue or any sort of thing like that burning off.

Anyway, do we know why the engine was offset?

The thrust puck is designed to (eventually) hold three Raptors, so none will be directly in the center. Test as you fly. Also gives them data on a single-engine landing if the other two were to fail.
